应用受体结合法检测胃泌素受体结合位点数,观察了脾虚大鼠胃粘膜胃泌素受体的变化规律与益气健脾中药黄芪的调控作用。结果显示大黄和利血平脾虚模型大鼠胃粘膜胃必素受体结合点数均有明显减少。黄芪在纠正脾虚证的部分病理证候的同时,对脾虚大鼠胃粘膜胃泌素受体结合位点数有显著上调作用,而对血清胃泌素水平却有一定降低作用趋势。高浓度黄芪注射液在体外可显著促进胃泌素受体结合。推测胃泌素受体参与了脾虚证的病理过程,黄芪健脾药理可能不是直接作用于胃泌素受体的内源性配基的结合部位,而有可能是一种类似酶别构效应机制的Ⅱ型受体激动剂。
The receptor-binding method was used to detect the gastrin receptor binding site number, and the changes of gastrin receptors in gastric mucosa of rats with spleen deficiency were observed and the regulating effects of Yiqi Jianpi Huangqi were observed. The results showed that the number of conjugated spots of gastric uremic receptors in gastric mucosa of rhubarb and reserpine spleen deficiency rats were all significantly reduced. While Huangqi is correcting some pathological syndromes of spleen deficiency syndrome, it has a significant up-regulation effect on gastrin receptor binding sites in gastric mucosa of spleen deficiency rats, but it has a decreasing effect on serum gastrin levels. High concentration of Astragalus injection can significantly promote gastrin receptor binding in vitro. It is speculated that gastrin receptor is involved in the pathological process of spleen deficiency syndrome. Huangpi Jianpi pharmacology may not directly affect the binding site of the endogenous ligand of gastrin receptor, but may be a similar allosteric effect mechanism. Type II receptor agonists.
