目的:研究不同剂量的镇肝熄风汤对自发性高血压大鼠(SHR)脂肪组织PPARγ2蛋白和PPARγ2mRNA表达的影响。方法:24周龄自发性高血压大鼠(SHR),雄性,50只,模型组、中药低剂量组、中药中剂量组、中药高剂量组、复方罗布麻组,每组10只,同源雄性WKY大鼠10只作为对照组,灌胃给药5周后,Western Blot方法测定脂肪组织PPARγ2蛋白表达,用Real-Time RT-PCR(Real-Time Quantitative RT-PCR)方法测定大鼠PPARγ2 mRNA基因表达。结果:各组与模型组比较,PPARγ2蛋白表达明显降低(P<0.01);各组与模型组比较,PPARγ2 mRNA表达明显降低(P<0.05);结论:镇肝熄风汤能减少脂肪组织PPARγ2蛋白表达和PPARγ2mRNA表达,从而减少成脂和保护心肌细胞功能。
Objective: To study the effect of different doses of Zhengan Xifeng Decoction on the expression of PPARγ2 protein and PPARγ2 mRNA in spontaneously hypertensive rats (SHR). Methods: 24-week-old spontaneously hypertensive rats (SHR), 50 male, model group, low dose Chinese medicine group, medium dose group, high dose group, compound Apocynum group, Ten male WKY rats were used as control group. The expression of PPARγ2 protein in adipose tissue was determined by Western Blot 5 weeks after intragastric administration. The expression of PPARγ2 mRNA in rat adipose tissue was detected by Real-Time RT-PCR gene expression. Results: Compared with the model group, the expression of PPARγ2 protein in each group was significantly decreased (P <0.01). Compared with the model group, the expression of PPARγ2 mRNA was significantly decreased (P <0.05). Conclusion: Zhengan Xifeng decoction can reduce the expression of PPARγ2 Protein expression and PPARγ2 mRNA expression, thereby reducing adipogenesis and protecting cardiomyocyte function.
