应用异型双功能连接剂SPDP将抗人肺腺癌单克隆抗体CMU15A与天花粉蛋白(TCS)交联,制备出新型免疫毒素。应用的MTT法检测其对人肺腺癌细胞系AGZY—83A的选择性杀伤活性。结果表明,CMU15A—TCS结合物具有高效靶向杀伤肺腺癌细胞能力,优于游离TCS及单抗与TCS混合物;与对非靶细胞的细胞毒作用相比较差异非常显著。提示该结合物是一种很有价值的免疫治疗剂,为今后的实验和临床应用提供了资料。
A new type of immunotoxin was prepared by cross-linking anti-human lung adenocarcinoma monoclonal antibody CMU15A and trichosanthin (TCS) with a heterotypic bifunctional linker SPDP. MTT assay was used to detect its selective killing activity on human lung adenocarcinoma cell line AGZY-83A. The results showed that the CMU15A-TCS conjugate has the ability to efficiently target lung adenocarcinoma cells and is superior to the free TCS and the mixture of the monoclonal antibody and TCS. The cytotoxicity against non-target cells is very different. Suggesting that the conjugate is a valuable immunotherapeutic agent for future experimental and clinical data.
