背景白藜芦醇是红葡萄酒中具有心血管保护作用的最重要的物质,可通过其强大的抗炎作用发挥抗动脉粥样硬化作用。目的观察白藜芦醇对因肿瘤坏死因子α(TNF-α)或sCD40L损伤的人脐静脉内皮细胞(HUVEC)的增殖活力和活性氧簇(ROS)的影响。方法体外培养HUVEC,白藜芦醇(1~50μmol/L)预孵育2h后,加入TNF-α(10μg/L)或sCD40L(10μg/L)刺激4h。用MMT法检测各组内皮细胞的活力;利用免疫荧光法检测各组HUVEC内活性氧的表达。结果TNF-α和sCD40L均可损伤HUVEC,使细胞增殖活力明显下降(分别为对照组的32.7%和26.0%,P<0.05),ROS表达增加(分别为对照组的3.4倍和4.9倍,P<0.05),而白藜芦醇(10、50μmol/L)可有效预防TNF-α和sCD40L对HUVEC的损伤(P<0.01)和抑制HUVEC内ROS的产生(P<0.01)。结论白藜芦醇可通过抑制HUVEC内ROS的产生,降低TNF-α或sCD40L对细胞的氧化损伤,发挥保护血管内皮的作用。
BACKGROUND Resveratrol is the most important cardiovascular protective agent in red wine and can exert antiatherogenic effects through its powerful anti-inflammatory action. Objective To observe the effect of resveratrol on proliferation activity and reactive oxygen species (ROS) of human umbilical vein endothelial cells (HUVEC) damaged by tumor necrosis factor-α (TNF-α) or sCD40L. Methods HUVEC and resveratrol (1~50μmol/L) were pre-incubated for 2h in vitro and then stimulated with TNF-α (10μg/L) or sCD40L (10μg/L) for 4h. The viability of endothelial cells in each group was detected by MMT assay; the expression of reactive oxygen species in HUVECs was detected by immunofluorescence. Results Both TNF-α and sCD40L could injure HUVEC, and the cell viability decreased significantly (32.7% and 26.0%, P<0.05, respectively) in the control group. The expression of ROS increased (3.4 and 4.9 times of the control group, respectively). <0.05), and resveratrol (10, 50μmol/L) can effectively prevent TNF-α and sCD40L damage to HUVEC (P <0.01) and inhibition of HUVEC ROS production (P <0.01). Conclusion Resveratrol can protect vascular endothelium by inhibiting the production of ROS in HUVEC and reducing the oxidative damage of TNF-α or sCD40L to cells.
