目的:观察阿仑磷酸钠联合辛伐他汀对体外成骨细胞、破骨细胞的影响,探讨2种药物联合应用治疗骨质疏松的可行性。方法:分离新生大鼠成骨细胞和破骨细胞,给予阿仑磷酸钠、辛伐他汀刺激,观察药物对成骨细胞增殖率、碱性磷酸酶活性及破骨细胞骨吸收的影响。结果:联合应用阿仑磷酸钠和辛伐他汀较单独用药更明显增强成骨细胞活性和抑制破骨细胞的骨吸收。结论:联合应用阿仑磷酸钠和辛伐他汀比单独应用阿仑磷酸钠可能对骨质疏松有更好的防治作用。
OBJECTIVE: To observe the effect of alendronate and simvastatin on osteoblasts and osteoclasts in vitro and to explore the feasibility of using two drugs in combination for the treatment of osteoporosis. Methods: Osteoblasts and osteoclasts were isolated from neonatal rats. Alendronate and simvastatin were used to stimulate osteoblast proliferation, alkaline phosphatase activity and osteoclast bone resorption. RESULTS: Combination of alendronate and simvastatin significantly enhanced osteoblast activity and inhibited osteoclastic bone resorption compared with single administration. Conclusions: Combination of alendronate and simvastatin may have a better preventive and therapeutic effect on osteoporosis than alendronate alone.
