目的 探讨STAT3的活化对霍奇金淋巴瘤 (HL)细胞凋亡的调控作用。方法 采用SP免疫组织化学方法检测了 4 5例不同亚型HL之RS/H细胞的STAT3、促凋亡基因fas和抗凋亡基因bcl- 2的表达。结果 在4 5例HL中 ,STAT3、fas及bcl- 2蛋白的阳性表达率分别为 6 4 4 % (2 9/ 4 5 )、6 2 2 % (2 8/ 4 5 )和 75 % (30 / 4 0 )。在不同亚型 (LP、NS、MC、及LD)中 ,STAT3的阳性表达强度无明显差异 (P >0 0 5 ) ;fas阳性强度依次降低 (P <0 0 5 ) ,而bcl- 2阳性强度逐渐增高 (P <0 0 5 )。统计学分析表明STAT3和fas两者的表达强度呈明显负相关 (γ= - 0 5 76 3,P <0 0 5 ) ,而STAT3和bcl- 2蛋白两者的阳性强度呈明显的正相关 (γ =0 6 35 7,P <0 0 5 )。结论 STAT3活化可能通过抑制细胞凋亡导致HL中RS/H细胞的恶性转化及高增殖性
Objective To investigate the role of STAT3 activation in the apoptosis of Hodgkin’s lymphoma (HL). Methods SP immunohistochemistry was used to detect STAT3, fas and anti-apoptotic gene bcl-2 in RS / H cells of 45 subtypes of HL. Results The positive rates of STAT3, fas and bcl-2 protein in 45 cases of HL were 62.4% (29/45), 62.2% (28/45) and 75% (30) respectively / 4 0). There was no significant difference in the positive expression of STAT3 in different subtypes (LP, NS, MC, and LD) (P> 0.05), followed by a decrease in fas positive (P <0 05) The intensity increased gradually (P <0 05). Statistical analysis showed that there was a significant negative correlation between the expression levels of STAT3 and fas (γ = - 0 5 76 3, P <0 05), while the positive expression of STAT3 and bcl-2 proteins was positively correlated γ = 0 6 35 7, P <0 0 5). Conclusion Activation of STAT3 may lead to malignant transformation of RS / H cells and hyperproliferation in HL by inhibiting apoptosis
