红霉素上调THP-1细胞组蛋自去乙酰化酶2表达减轻氧化应激下的糖皮质激素抵抗

目的探讨红霉素(EM)对氧化应激下人THP-1单核细胞糖皮质激素抵抗的作用及机制.方法 EM预孵育THP-1细胞后予烟草烟雾提取物(CSE)刺激细胞,用质粒脂质体法将构建好的组蛋去乙酰化酶2沉默RNA (HDAC2-siRNA)转染细胞,ELISA检测细胞培养上清白细胞介素8 (IL-8)的含量,实时荧光定量PCR (qRT-PCR)和Western blot法分析HDAC2表达情况.

目的通过EGFR特异性单链抗体介导siRNA内化入TKIs耐药肿瘤细胞,恢复药物敏感性.方法 1、单链抗体的设计及纯化:设计EGFR特异性scFv/s-9R核酸表达序列并全序列化学合成后连接于表达载体,经酶切鉴定及测序后转化大肠杆菌BL21(DE3).将scFv、s-9R的单克隆工程菌于37℃培养至OD600约为0.4-0.6时,添加异丙基-β-D-硫代半乳糖苷(IPTG)诱导表达.
