脂联素介导ciRS-2439-5p调控牛前体脂肪细胞分化

  CircRNA在生命活动中发挥重要作用,但是到目前为止,circRNA调控脂肪代谢的机制并不清楚.本研究以郏县红牛心、肝、脾、肺、肾、肌肉和脂肪共7个组织的circRNA-mRNA联合测序为基础,利用权重共表达分析(WGCNA Analysis),生物信息学分析和circRNA-mRNA组学贯穿分析筛选到4条与脂肪发育密切相关的组织特异表达circRNA,分别命名为ciRS- TMEM165、ciRS-2439-5p、ciRS-FGF18和ciRS- PPHLN1.前三条circRNA通过ADIPOQ协同调控pparG信号通路,其中ciRS-2439-5p对牛前体脂肪细胞分化的影响,及其与ADIPOQ的共表达关系已通过超表达和实时定量PCR实验所证明.总之,ciRS-2439-5p极有可能是影响脂肪代谢的重要circRNA,进一步分析它发挥作用的信号机制是我们下一步研究的重点.
