采用高效液相色谱、电喷雾多级串联质谱及微量核磁共振技术对2',3',5'-三氧乙酰基-N6-(3-羟苯胺)腺苷在大鼠体内尿液中的代谢物进行结构解析

2,3,5-三氧乙酰-N6-(3-羟苯胺)腺苷(图1)是一种新型的腺苷类似物,体内外实验证明其具有显著的调血脂活性[1].本实验是采用高效液相色谱(HPLC)、质谱(MS)及微量核磁共振(microprobeNMR)技术[2]对其在大鼠尿液中的代谢物进行结构解析.利用HPLC分离、富集尿样中的代谢物,采用微量探头,采集了7个主要代谢物的1H、13C、2DNMR谱(图2),结合质谱信息,得出它们分别为Ⅰ相代谢产物2,3,5-三羟基-N6-(3-羟苯胺)腺苷、N6-(3-羟苯胺)腺嘌呤、N8-羟-N6-(3-羟苯胺)腺嘌呤,Ⅱ相代谢产物N6-(3-O-β-D-葡萄糖醛酸基苯胺)腺嘌呤、N8-羟基-N6-(3-磺酸基苯胺)腺嘌呤、2,3,5-三羟基-N6-(3-O-β-D-葡萄糖醛酸基苯胺)腺苷及2,3,5-三羟基-N6-(3-磺酸基苯胺)腺苷.
