The non-classical HLA class 1 antigen HLA-G expression was firstly observed to be restricted to the fetal-maternal interface on the extravillous cytotrophoblasts and proposed to play important roles in the maintenance of fetal-maternal immune tolerance.However, under pathophysiological conditions, HLA-G has been correlated with the immune escape of tumor cell and virus-infected cells, and with the acceptance of transplants, tn the scenario of malignancies, roles of aberrant HLAG expression have been extensively investigated and its clinical relevance has been postulated.We will summarize HLA-G biology and how it participates in tumor cell immune escaping, metastasis and disease progression.Special attention will be paid to its role as a diagnostic biological marker and also as a therapeutic target in cancers.
