Heat shock protein 90(Hsp90) is a member of the heat shock protein family that stabilizes a variety of proteins required for survival of cancer cells.Recently,a number of Hsp90 inhibitors are under clinical trials for cancers chemotherapy.In addition to the preceding report,we also discovered two additional classes of Hsp90inhibitors bearing the core structure of 1,2,3-triazolo[1,5-a]pyrazine-6-one and 1,2-isoxazolo[4,3-c]pyridine-6-one,respectively.All the compounds are synthetically accessible by using intramolecular [3+2] cycloaddition as key step,and showed potent anticancer activities in an array of cancer cell lines.
