Up to date, small molecules that can selectively target cancer stem cells are still very rare, and basically have no common structural feature.We discovered a series of compounds that can selectively ablate AML stem/progenitor cells, and then established the first preliminary SAR against AML stem/progenitor cells.In ex-vivo experiment with NOD/SCID mice models, the survived primary AML cells after treatment with these compounds demonstrated significantly lower engraftment in mice than the equal amount of saline treated primary AML cells.Drug candidate ACT001, demonstrates excellent therapeutic effect when doses orally, and is well-tolerated in acute and chronic toxicity study.The broad therapeutic index indicates that ACT001 is a promising anti-cancer drug candidate for clinical trial.
