  CRISPR/Cas9是细菌和古细菌在长期演化过程中形成的一种适应性免疫防御,可用来对抗入侵的病毒及外源DNA.crRNA通过碱基配对与tracrRNA (trans-activating RNA)结合形成tracrRNA/crRNA复合物(gRNA),此复合物可引导Cas9核酸酶在与crRNA配对的序列靶位点造成DNA双链断裂.目前通过人工改造与设计RNA,理论上我们可以知道Cas9对大多数DNA特异序列的定点切割.
