Objective To evaluate the effect of co-expression DNA vaccine containing GAD65 fragment and IL-10 gene in preventing young nonobese diabetic (NOD) mice from developing autoimmune diabetic mellitus and observe its modulation on Treg cells dynamically.Method The plasmid co-encoding GAD65190-315 fragment and IL-10 gene was used as the DNA vaccine.Female NOD mice 4 weeks old were randomly divided into 2 groups: the GAD65/IL-10 vaccine group(n=36) and the pBudCE4.1 vector control group(n=38) by intramuscularly injected with 75ug plasmid once a week for two consecutive weeks.The accumulative diabetes incidence was followed up to 30 weeks of age in each group.Insulitis was evaluated with Hematein Eosin stained pancreatic sections from 12-week old NOD mice.CD4+CD25+Foxp3+(Tregs)lymphocytes in spleens and pancreatic draining lymph nodes in each group were measured by flow cytometry.
