To investigate whether MyD88 in the gut epithelial brush border plays a role in obesity and metabolic syndrome, we studied a transgenic mouse line expressing MyD88 only in villus (VillinMyD88+/+MyD88-/-mice).In this study, we fed young male wild type B6 (n=8),MyD88-/-B6 (n=6) and VillinMyD88+/+MyD88-/-B6 mice (n=13) with high fat diet (HFD) for 16 weeks.We investigated the metabolic, immunological and gut bacteria profiles of these mice.We found that VillinMyD88+/+MyD88-/-B6 mice had lower body weight, net weight gain, total body fat mass, white adipose tissue weight, and smaller adipocytes compared to B6 and/or MyD88-/-B6 mice (P<0.05).Glucose and insulin tolerance tests showed that VillinMyD88+/+MyD88-/-B6 mice were more insulin sensitive than B6 and MyD88-/-B6 mice (P<0.05).
