Poly(ADP-ribose) polymerase-1 (PARP-1) plays important roles in multiple DNA damage response pathways rendering it a promising target in cancer therapy.However, cytoprotective effects of PARP1 inhibition (P-1I) may limit this potentiality.We investigated effect of P-lI on modulation ofkinase cascades and regulation of transcription factors in a rodent septic shock and cellular oxidative stress models.We demonstrated at the first time P-1I activates Akt and induces MKP-1 expression thereby down-regulates MAPKs resulting in mitochondrial-and cytoprotection.
