Single domain antibodies (sdAbs) are antibody fragments consisting of only one protein domain.A llama sdAb phage display library was screened against non-small cell lung carcinoma cells A549 and an sdAb,AFAI,was identified.AFAI was pentamerized to become a pentabody ES1,which is found to be highly specific to large cell lung carcinoma and lung adenocarcinoma with about 97% specificity.Its antigen was identified as carcinoembryonic antigen-related adhesion molecule 6 (CEACAM6).CEACAM6 was known as a marker for various tumors especially colon carcinoma.Surprisingly,AFAI has higher specificity to lung carcinomas than to colon ones,indicating the importance of epitope in biomarker discovery.Another EGFR-specific sdAb EG2 was fused to human Fc to become a chimeric heavy chain antibody EG2-hFc.Using microPET technology,EG2-hFc was shown to have excellent tumor targeting ability.These results demonstrate that sdAb is a good tumor antigen discovery tool as well as a good candidate for antibody drags.
