Laser speckle contrast imaging(LSCI)combined with skin optical clearing method makes it possible to image cutaneous microcirculation with high resolution and contrast.But it has not been investigated whether it is competent to monitor skin blood flow dynamics.The purpose of this work is to evaluate the capacity of the combined method to monitor cutaneous arteriovenous blood flow dynamical response to vasoactive noradrenaline.The results indicated that optical clearing method induced no noticeable changes in cutaneous microvasculature; the dynamic changes in vascular diameter and flow velocity caused by drug can be easily monitored using LSCI with superior sensitivity; the distinct dynamical response time and amplitude between arteries and veins can be assessed by the rebuilt arteries-veins separation through the transparent cutaneous microvasculature.On the contrary,it was difficult to detect the skin blood flow dynamics and these differences among vessels with diverse kind and size through the intact skin.Thus,skin optical clearing method not only shows a considerable value in enhancing the capacity of LSCI to investigate skin blood flow dynamics with higher resolution,contrast and sensitivity,but also makes it available to noninvasively assess cutaneous arteriovenous pharmacological intervention and treatment.
