Five days after germination, mungbean seedlings were used as explants after the removal of primary roots.The explants were treated with different concentrations of CaCl2, ruthenium red (an inhibitor of the endomembrane Ca2+ permeable channel), EGTA (a chelator of extracellular Ca2+), CaCl2 + ruthenium red and CaCl2 + EGTA.The number and fresh weight of adventitious roots were quantified after 5 d of treatments meanwhile incubated at 25 ± 1℃ in 14 h photoperiod as during germination.When compared with the controls, 5 mM CaCl2 treatments significantly increased the number and fresh weight of adventitious roots per explant, however, 1-10 μM ruthenium red or 0.5-1 mM EGTA strongly decreased the number and fresh weight of adventitious roots per explant and the inhibitory effects were dose-dependent.When compared with the treatments of EGTA or ruthenium red alone, the treatments with 5 mM CaCl2 together with 0.5 mM EGTA or 5 mM CaCl2 together with 1 μM ruthenium red can significantly reversed the number and fresh weight of adventitious roots per explant, indicating that the inhibitory effects of EGTA or ruthenium red on adventitious rooting were eliminated by 5mM CaCl2.The results obtained from the experiments showed that Ca2+, especially the cytosolic Ca2+, acting as a signal molecule, play an important role during adventitious rooting of plants.
