Nijmegen breakage syndrome (NBS) as well as Ataxia Telangiectasia (A-T) is a radiation-hypersensitive genetic disorder, showing chromosomal instability, radioresistant DNA synthesis, and predisposition to malignances.The product of responsible gene, NBS1 forms a complex with MRE11 and RAD50 (MRN complex).NBS1 also interacts with ATM upon generation of DNA double-strand breaks (DSBs), and this interaction is indispensable for sufficient ATM activation.So far, we identified other its interactions, with RNF20 (ubiquitin ligase for H2B), or Rad18 (ubiquitin ligase for PCNA).
