金黄色葡萄球菌(Staphylococcus aureus)是人类的一种常见病原体,由于抗生素的滥用使得金黄色葡萄球菌产生多重耐药性.本研究发现异硫氰酸苄酯对标准金黄色葡萄球菌菌株ATCC25923 和临床上MRSA 菌株表现出良好的杀菌效果,MIC 均为10 μg/mL.通过扫描电镜观察发现异硫氰酸苄酯导致金黄色葡萄球菌细胞发生皱缩损伤.使用碘化丙啶(PI)荧光探针标记,结合流式细胞仪分析确定异硫氰酸苄酯可以杀死金黄色葡萄球菌细胞.通过转录组测序、WGCNA 分析以及生物信息学分析等确定SmpB 作为异硫氰酸苄酯诱导金黄色葡萄球菌死亡的枢纽基因.通过蛋白质体外表达、等温滴定量热法及Western blot 杂交等验证并明确了异硫氰酸苄酯与SmpB 互作位点.通过构建金黄色葡萄球菌SmpB 突变体以及分析下游调控因子表达情况,揭示了异硫氰酸苄酯通过靶向SmpB 诱导金黄色葡萄球菌死亡的分子机制.分析结果显示异硫氰酸苄酯可特异性靶向结合SmpB,影响反式翻译,诱导ClpX 蛋白酶系统启动细胞死亡进程.该研究为异硫氰酸苄酯控制金黄色葡萄球菌污染提供了理论指导,为新型杀菌剂的开发提供了靶标.
