In this paper, we have designed two kinds of schemes to prepare aspirin and esomeprazole magnesium compound enteric coated pellet capsule which can inhibit gastric acid secretion by esomeprazole magnesium and antagonize side effects of aspirin on gastrointestinal.Thena better antithrombotic treatment will be achieved.The first scheme:To begin with, the aspirin pellets are prepared by extrusion spheronization;Then the esomeprazole magnesium pellets are prepared by fluidized bed coating irrigation method;Finally, the aspirin and esomeprazole magnesium enteric coated pellets are prepared by fluidized bed coating method and the two kinds of enteric coated pelletsare filled in the same capsules after content determination to form compound capsules;the second scheme: Firstly,the aspirin pellets are prepared by extrusion spheronization;Then the surface of aspirin pellets are coated with isolation layer I by fluid bed coating method , then package esomeprazole magnesium layer, and then package isolation layer H.Finally, an enteric layer.Aspirin and esomeprazole magnesium are contained in the same pellets of compound capsules.In vitro test results showed that, the release rate of two kinds of compound capsuleswas less than 10%in 0.1mol/L hydrochloric acid solution in 2h.;the release rate of aspirin and esomeprazole magnesiumin the pH6.8 PBS were 5.9% and 78.5% in 15min,77.4% and 83.5 in 60min, respectively.The f2 among groups was greater than 50, indicating that the formulation and process have good reproducibility;in vivo results showed that, for two kinds of compound capsule preparations, the Tm ax of esomeprazole magnesium was 1.5h after administration.However, the TM ax of aspirin was 3.0h for the first scheme and 4.0h for the second scheme, respectively.For the second compound capsules, esomeprazole magnesium release firstly, inhibiting gastric acid secretion, and protecting the gastric mucosa,to avoid side effects of aspirin.So the second scheme can preferablyexertthe advantages of compound preparation, which provides a new Choice, new idea for the development of compound preparations.
