目的:探讨壳聚糖对镉染毒大鼠肝脏线粒体氧化损伤的拮抗作用.方法:将40 只清洁级健康SD 大鼠随机分为5 组,分别为对照组、0.8mg/kg 氯化镉组和20、100、500 mg/kg 壳聚糖+0.8 mg/kg 氯化镉组,每组8 只,雌雄各半.壳聚糖采用灌胃方式;2h 后,采用腹腔注射方式染毒氯化镉,每天1 次,连续染毒7 d.对照组和0.8 mg/kg 氯化镉组灌胃蒸馏水,对照组腹腔注射生理盐水.测定大鼠肝脏线粒体内超氧化物歧化酶(superoxide dismutase,SOD)比活,还原型谷胱甘肽(glutathione,GSH)、细胞色素C(CytC)及丙二醛(MDA)含量.结果:与对照组比较,0.8 mg/kg CdCl2 组肝脏脏器系数升高,差异具有统计学意义(P<0.05);与0.8 mg/kg CdCl2 组比较,100 mg/kg、500 mg/kg 壳聚糖+0.8mg/kg CdCl2 组肝脏脏器系数均降低,差异有统计学意义(P<0.05).0.8 mg/kg氯化镉组大鼠肝线粒体SOD 比活及GSH、CytC 含量均下降,MDA 含量明显升高,与对照组比较,差异有统计学意义(P<0.05);与0.8 mg/kg 氯化镉组比较,20、100、500 mg/kg 壳聚糖+0.8 mg/kg 氯化镉组大鼠肝线粒体SOD 比活均升高,500mg/kg 壳聚糖+0.8 mg/kg 氯化镉组CytC 含量升高,MDA 含量下降,差异有统计学意义(P<0.05);各剂量壳聚糖+0.8 mg/kg 氯化镉组大鼠肝脏线粒体GSH含量均无明显变化.结论:壳聚糖对镉致大鼠肝脏线粒体氧化损伤具有一定的拮抗作用.
