The rapidly emerging field of synthetic biology demands new,efficient,and economical methods for synthesis of artificial genes,operons,and even genomes.In many cases the traditional methods for obtaining functional DNA sequences through cloning are not applicable due to the novelty of genetic material.Many companies offer gene synthesis service,but the price range is still too high for most molecular biology labs.We have proposed a novel long DNA synthesis method based on USER technology [1].The method consists of:i) synthesis of building blocks up to 500 bp; ii) assembly of genes up to 5 kb; iii) error correction reassembly; iv) assembly of operons up to 30 kb.The advantages of this method include the minimization of restriction endonuclease digestion and ligation steps,the ability to produce long DNA sequences in very short time (1-3 weeks),and it is easy to use in any molecular biology laboratory.
