Treatment of patients with metastatic soft-tissue and bone sarcomas remains a significant challenge with few new options.The kinase inhibitors imatinib and sunitinib have helped patients with gastrointestinal stromal tumors (not further discussed here).Trabectedin and mifamurtide are the only recently approved drugs (in Europe) for patients with certain types of sarcoma.The mTOR pathway, which controls protein translation and cell growth and metabolism, is commonly dysregulated in cancerl.Deforolimus, a non-prodrug analog of rapamycin, is a potent mTOR inhibitor and has been evaluated clinically as iv and oral formulations, and has shown promising activity in a broad range of sarcomas.
