Objective: In order to enhance lignin degradation efficiency of a lignin degradation bacterial consortium named LDC,its culture condition and medium composition were optimized.Method: a single-factor experiment and the response surface method of Box-Behnken Design were used to optimize the culture condition of LDC,a single-factor experiment and an orthogonal test were employed to optimize the carbon and nitrogen source.Result: The optimal culture condition of LDC:the culture temperature,initial pH and dissolved oxygen were 32.0 ℃,8.0 and 7.8 mg/L respectively.The best carbon and nitrogen source were composed of soluble starch 5.0 g/L,sodium nitrate 2.0 g/L and ammonium nitratencreased 1.0 g/L.Under above conditions,the lignin in reeds was degraded 70.0%by LDC.Conclusion: The growth cycle was shorted by 50%and the lignin degradation rate was increased by 14.8%in reeds by LDC.The high lignin degradation efficiency of LDC applied bases for bio-pulping.
