目的:研究活性氧簇(Reative oxygen species,ROS)在严重烧伤后肺损伤中对肺微血管内皮细胞的作用。方法 将成年雄性SD大鼠按照随机数字表法分为正常对照组(3只)、烫伤组(25只),烫伤组大鼠于恒温水烫仪中用95℃热水浸浴背部18S造成30% TBSA Ⅲ度烫伤,正常对照组大鼠用37℃水浴模拟致伤。烫伤组分别在烫伤后6h,12h,24h,36h,48h,60h,72h腹主动脉取血并分离血清,ELISA检测血清中ROS水平;组织块法培养原代培养肺微血管内皮细胞(pulmonary microvascular endothelial cells,PMVECs),用兔抗大鼠Ⅷ因子抗原多克隆抗体免疫荧光鉴定。
