目的 探讨重组人粒细胞巨噬细胞刺激因子(rhGM-CSF)不同时机应用对于小鼠深Ⅱ度烫伤创面的影响.方法 (1)取24只体质量为(20.0±1.0)g的雄性BALB/c小鼠,建立小鼠深Ⅱ度烫伤创面.按照随机数字表法分为伤后第1天使用rhGM-CSF组、伤后第3天使用rhGM-CSF组、伤后5天使用rhGM-CSF组,伤后1周使用rhGM-CSF组.rhGM-CSF局部外用共计一周.于伤后5天、10天分别切取3只各处理因素间小鼠皮肤样本,实时定量RT-PCR和蛋白质印记法检测VEGF、Ⅰ、Ⅲ型胶原表达情况.(2)另取20只BALB/c小鼠,按照前述方法 分组并处理,于伤后1天、5天、7天、14天观察不同组别的创面愈合时间,检测创面愈合率.对数据行单因素方差分析.结果 (1)伤后3天使用rhGM-CSF组VEGF、Ⅰ、Ⅲ型胶原mRNA表达水平与其余各组比较均升高,且差异有统计学意义(p<0.05).伤后3天使用rhGMCSF组VEGF、Ⅰ、Ⅲ型胶原蛋白表达水平与其余各组比较均显著升高(p<0.05).(3)伤后3天使用rhGM-CSF组创面愈合时间明显缩短,该组在伤后第14天创面愈合率明显高于其余各组.结论 小鼠深Ⅱ度烫伤创面于伤后第3天使用rhGM-CSF,与其他各组相比,能够明显加速创面愈合.
