Objective: To establish a quantification method for quercetin,rutoside, kaempferol in Ilex hainanensis by HPLC.Methods : RP-HPLC is applied on the determination of quercetin,rutoside, kaempferol in Ilex hainanensis with C 18 column(4.6 mm×250 mm, 5 μm),DAD detector, and a mixture of acetonitrile-0.2% phosphoricacid (60∶ 40,Ⅴ/Ⅴ) as a mobile phase with the flow rate at 1.0mL·min-1.Three flavonoids were detected at251 nm, column temperature was set at 35℃.Result:.Quercetin, rutoside and kaempferol were linear in the range of0.420~8.38 μg, 0.168~3.36 μg and 0.075~ 1.50 μg, The average recovery of rutoside, quercetin and kaempferol was 99.65%, 99.27%, 98.86% and RSD(n=5) was 0.47% ,2.04% ,1.82% (n=5) respectively.Conclution : The method is accurate,sensitive and can be used rapidly for content determinatinon of rutoside, quercetin and kaempferol from Ilex hainanensis.It can be applied to the quality control of this crude drug.
