The effect of Broccoli extract on proliferation and apoptosis in colon cancer cell line SW620

  To study the inhibitory effects of broccoli in colon cancer cells line SW620 and its possible mechanism.SW620 cells were treated with broccoli extract in different concentration.In order to further explore the anti-tumor mechanisms of broccoli, with different concentration broccoli crude extract processing colon cancer cell line SW620, using MTT and drawing growth curve, this experimental investigates the SW620 cell proliferation condition.The transcription and expression level change of SW620 of apoptosis related gene p53 and caspase-3 were detected by q-PCR, analysis the influence mechanisms of broccoli crude extract on SW620 cell proliferation and apoptosis.The results showed that different concentrations of broccoli crude extract has obvious inhibition effect on SW620 cell proliferation which increases obviously with the increasing concentration, and the highest inhibition rate reaches as high as 90.1%.By MTT method IC50 of broccoli crude extract has calculated as 0.1 mg/mL.After processing by the broccoli crude extract on IC50, the following changes of SW620 cell has occurred: the growth of cells is obviously restrained, cell falls off, deformation and adherent ability drops, appearing the phenomenon of apoptosis.Cell genome DNA banding appears obviously tailing phenomenon, the transcriptional level of caspase-3 detected by fq-PCR compared with the control group has increased more than three times, moreover, the same level of p53 has increased dozens of times.The research results show that the broccoli crude extract can inhibit the growth of colon cancer cell line SW620, and its inhibition mechanism may be show that the active material induced the up-regulated expression of p53 and caspase-3 those two important apoptosis regulation gene in apoptosis pathway, leading the cell apoptosis.
