A genetic linkage map of Silurus meridionalis (Teleostei: Siluriformes)derived from RAD sequencing

  Genetic linkage maps are indispensable tools in genetic and genomic studies.The newly developed highresolution genetic map can provide valuable genetic information for the assembly of genome sequences, comparative analyses of synteny, dissection of complex genetic traits.Recently, the development of restriction-site associated DNA (PAD) method holds great promise for constructing high-resolution linkage maps in model and non-model organisms.Here, we constructed a genetic linkage map of Silurus meridionalis based on 29 116 SNPs using RAD-seq, which to our knowledge had the highest number of markers among aquaculture species.The total length of the genetic linkage map was 24 121.567 cM with an average marker interval of 0.83 cM.All of the SNPs are distributed across all 29 linkage groups.However, we found a large interval from the bottom 18 markers in LG6 which illustrates there is a centromeric region in this linkage group.In this study, we used RAD-seq to produce a high quality linkage map of Silurus meridionalis.It demonstrates that high-resolution genetic map can provide a basis for future genetic and genomic studies and RAD markers should be a useful molecular tool for a range of non-model organisms.
