谭小艳等从广西合浦县柑橘叶片表面分离获得一株对柑橘溃疡病菌(Xanthomonas axonopodis pv.citri)具有较强拮抗作用的枯草芽孢杆菌菌株Bv10.研究发现,除Xanthomonas spp.外,菌株Bv10对多种植物病原真菌也有不同程度的抗菌活性.为深入了解菌株Bv10抗菌活性的物质基础,作者以芝麻白绢病菌Athelia rolfsii作为指示菌,对菌株Bv10产生的胞外抗菌物质进行了纯化与鉴定.经盐酸沉淀、甲醇萃取以及反相高效液相色谱(RP-HPLC)等方法,从菌株Bv10的无菌发酵液中提取和纯化到A和B两种对芝麻白绢病菌具有较强抑制作用的活性物质,其分子量分别为1 042Da和1 057Da.纯化物A和B浓度都为200mg/L时可以完全抑制芝麻白绢病菌菌核的萌发;当浓度为100mg/L和50mg/L时,纯化物A对菌核萌发的抑制率分别为82.2%和73.3%,纯化物B对菌核萌发的抑制率分别为77.8%和68.9%.纯化抗菌物质A和B对芝麻白绢病菌的抑菌中浓度EC50值分别为36.7866mg/L和43.0329mg/L.显微镜观察结果表明,芝麻白绢病菌在菌株Bv10抗菌物质的作用下,菌丝体扭曲变形,生长紊乱,分隔处缢缩,甚至在分枝处断裂,菌丝顶端膨大,原生质体凝聚,顶端成囊泡状.MALDI-TOF-MS/MS CID质谱分析两种纯化物分子的一级结构均为Pro-Asn-Ser-βAA-Asn-Tyr-Asn-Gln,将纯化物A鉴定为脂肽化合物伊枯草菌素iturin A2,将纯化物B鉴定为iturinA4.胰蛋白酶、胃蛋白酶、木瓜蛋白酶、果胶酶对纯化物的抗菌活性也没有影响.
