Objective: To determine the function of IL-9 in the development of CLL through STAT3, STAT6/miR-155, and miR-21 pathways.Methods: Data of patients who were diagnosed with CLL at Shandong Provincial Hospital from 2010 January to 2012July were collected.All patients were untreated, and their lymphocytes exceeded 90%.We measured the serum levels of IL-9 in 47 patients with CLL and healthy controls using ELISA.Expression of pSTAT3 and pSTAT6 in the peripheral blood mononuclear cells (PBMCs) from 20 CLL patients were detected using Western blot, with upregulation of IL-9and B cells from healthy samples.MiRNA155 and miRNA21expression in 20 patient and healthy samples was analyzed using real-time quantitative PCR.IL-9 production was determined by Western blot and real-time quantitative PCR in CLL and miR-transfected CLL cells, which were treated with IL-9 at various time points.These cells were then exposed to WP1066 or A77-1726 for 48 h to detect the IL-9 secretion.Cell proliferation was finally observed using CCK-8 incorporation assay.Cell apoptosis was investigated using annexinⅤ-FITC/PI staining followed by flow cytometry.
