SKP2, an F-box protein targets cell cycle regulators including cycle-dependent kinase inhibitor p27KIP1 via ubiquitinmediated degradation.SKP2 is frequently overexpressed in variety of cancers.We investigated the role of SKP2 and its ubiquitin-proteasome pathway in colorectal carcinoma using a panel of cell lines and clinical samples and SCID mouse model.Using immunohistochemistry on 448 samples, our data shows that expression of SKP2 is inversely associated with p27kip 1 protein levels (p0.0001) and patients with high level of SKP2 and low level of p27kip 1 have decreased overall survival (p=0.0057).Our in vitro study show that treatment of colorectal carcinoma cell lines (Colo320, HCT-15 and SW480) with PS341, an inhibitor of proteasomal pathway suppressed growth and induces apoptosis in all cell lines.
