目的:观察米非司酮(mifepristone,MIF)对慢性应激大鼠脂代谢和肝脏中GRmRNA、PPAR-γmRNA表达的影响.
方法:采用高脂饲料联合四种不同的应激方式(悬尾30 min;PVC筒固定2h;纱布束缚2h;大鼠固定架制动2h)建立慢性应激大鼠模型.随机分为正常对照(control)组、应激模型(model)组、米非司酮-低剂量(MIF-L)(10 mg·kg-1·d-1)、中剂量(MIF-M)(25mg·kg-1·d-1)、高剂量(MIF-H)(50 mg·kg-1·d-1)组,正常组和模型组给予等量蒸馏水.每周测定每组大鼠平均饮水量、摄食量、大鼠体重变化,给药4周后断头处死,测定血清中血脂四项(HDL-C、LDL-C、TG、TC)水平,HPA轴激素(CRH、ACTH、CORT)水平,采用Real Time PCR测定大鼠肝脏组织中GRmRNA,PPAR-ymRNA的表达.
结果:相比control组,model组TG、TC、LDL-C、CRH、ACTH、CORT升高;与model组比,MIF-L降低TC,升高HDL-C,MIF-M、MIF-H两组对TC、TG均有改善作用,且差异有显著性(P<0.04,P<0.05),MIF-M降低LDL-C;MIF降低CRH、ACTH水平,升高CORT水平,且MIF-H组CORT升高差异有显著性(P<0.01).
结论:MIF改善应激大鼠脂代谢紊乱,其有可能是通过升高肝脏中GRmRNA和PPAR-ymRNA的表达发挥作用.
