Protein arrays have emerged as a technology to study protein expression and protein function in a high-content and high-throughput manner.One obvious application of protein arrays is to profile protein expression, auto-antibody production and glycosylation in patients specimen.Through identification of unique biomarkers or biosignatures, protein arrays may have great impact on predictive and personalized medicine.Four different approaches of protein arrays including sandwich-based antibody arrays, label-based antibody arrays, quantitative antibody arrays and enhanced protein profiling arrays have been developed to detect protein expression.These innovative technologies have been integrated into our biomarker discovery program.The expression levels of 174 cytokines were determined using antibody arrays in ovarian cancer patients, benign ovarian subjects and normal individuals.Our data showed a group of cytokine expression levels were significantly changed in ovarian cancer patients and benign subjects.Neural network analysis revealed a specific cytokine expression profile among ovarian cancer, benign tumor and normal subject.Our study suggests that protein arrays hold great promising in biomarker discovery.
