This paper describes an efficient colchicine-mediated technique for the in vitro induction of octoploids in Pogostemon cablin and its confirmation by flow cytometry and chromosome count.The optimum concentration of colchicine treatment for octoploid induction was 0.05% for 72 h.The chromosome number of octoploid seedlings was 2n = 8x = 128.Colchicine-induced tetraploids remained stable after 6 months in soil.33 lines of autooctoploid plants were obtained.Colchicine-induced octoploids remained stable after 6 months in soil.Leaf characteristics of tetraploids and octoploids in P.cablin were compared.It was determined that leaf sizes and stomatal sizes of transplants were suitable parameters for identifying putative octoploids in P.cablin.
