Objective: To clone the 1-deoxy-D-xylulose-5-phosphate synthase cDNAs from Alpinia officinarum.Methods: The DXS cDNAs were cloned by homology-based RT-PCR and RACE methods.The characterization of the cDNA sequences and the deduced protein sequences were analyzed bioinformaticly.Result: Two full-length cDNAs encoding DXS were cloned from the rhizome of A.officinarum,The new cDNAs were designated as AoDXS1 and AoDXS2 with a 2148 bp and a 2136 bp open reading frame respectively,encoding polypeptides of 715 and 711 amino acids with calculated molecular mass of 77.23 and 51.48 KDa respectively.Bioinformatic analyses revealed that AoDXS1 and AoDXS2 showed extensive homology with DXSs from other plant species.Conclusion: The deduced polypeptides of AoDXS1 and AoDXS2 belonged to class Ⅰ and classⅡ DXS respectively.
