Ovarian cancer patients show increased levels of lysophosphatidic acid (LPA) in their serum and ascetic fluid.Using DNA-protein interaction array, our studies have identified novel pathways through which LPA can promote the proliferation of ovarian cancer cells via G12-family of G proteins.In contrast, ovarian cancer cell metastasis appears to involve both the G12-as well as Gi-family of G proteins.In addition, we unravel a novel role for LPA in promoting an acute metabolic reprogramming in ovarian cancer cells.Furthermore, we demonstrate that at least one node of this signaling pathway can be inhibited by Thymoquinone.Thus, our studies establish further that LPAstimulated signaling nodes can be targeted for cancer prevention and chemotherapy.This work was supported by grants from the National Institutes of Health (CA123233, CA 116984, and GM 103639).
