寡糖链非还原端单糖种类测定初探

  活性多糖或寡糖通过作用于巨噬细胞(Mφ)膜上的MR 受体,使其释放某些前炎症因子或细胞因子,对机体产生免疫调节作用。目前,多糖或寡糖免疫活性的筛选主要是通过活体动物及细胞药理学实验进行,操作复杂、成本高、周期长。基于MR 受体能特异性地识别端基为甘露糖、岩藻糖及N-乙酰葡萄糖胺的糖链,我们尝试建立一种基于常规仪器分析的寡糖初筛方法,从寡糖端基单糖组成入手,建立了一种端基单糖种类测定方法,快速对具有免疫调节活性的糖类物质进行筛选,根据降解后端基单糖(非还原端)与其它位置单糖甲基化程度的差异,识别非还原端基单糖。
