里氏木霉FS10-C基质筛选及固体发酵条件研究

  里氏木霉(Trichoderma reesei)FS10-C由本课题组前期研究从土壤中筛选而得,该菌株在Cu浓度为600 mg·L-1,Cd浓度为300 mg·L-1时仍能生长,并且能强化海州香薷和伴矿景天对铜镉污染土壤的吸取修复.因此,为扩大其应用前景,本研究以廉价的农副产品作为原料,对木霉FS10-C固体发酵培养基进行了筛选,并优化发酵条件,简化生产工艺,为将其成功开发为土壤重金属污染修复的生物修复剂提供参考.研究表明,木霉菌在单一麦麸基质上菌丝生长旺盛;在单一桔皮基质上产孢量高,麦麸与桔皮的混合物则表现出了对FS10-C菌生长的促进功能,产孢量达到9.47×109个·g-1,是单一桔皮的2.08倍.以基质配比、接种量、含水量和培养时间为试验因素,分别取3个水平,按[L9(34)]正交表进行正交试验,确定了重金属耐性木霉FS10-C平皿固体发酵的最佳条件为:桔皮、麦麸按1∶1比例配伍,固水比为1∶1.2,接种量为5%,发酵温度为28℃,发酵14d后产孢量可高达2.43×1010个·g-1.为进一步验证发酵效果,将其应用于50L固体发酵罐,各参数设置为:固水比1∶0.8;物料装量6~10 kg;通气量20~30 L·min-1,压力0.03~0.05 Mpa,接种量5%,温度28℃,发酵10d后产孢量达到1.44×1010个·g-1.由此可见,采用本研究确定的固体发酵条件可实现木霉FS10-C的高密度培养;桔皮与麦麸混合物不失为一良好培养基质.
