  Objective Using laser-Doppler flowmetry (LDF),the influence of tetramethylpyrazine (TMP) on regional cerebral blood flow (rCBF) of temporal cortex in rat after focal cerebral ischemia/reperfusion injury was monitored dynamically.Methods Male SD rats were randomly divided into sham-operation group,ischemia group and TMP treated group with different dose (10 mg/kg,20 mg/kg,40 mg/kg,or 80 mg/kg).Perfusion Units (PU),which is the main index of rCBF,Concentration of Moving Blood Cells (CMBC),Velocity (V) and Total Backscatter (TB) as the observation indexes of LDF were continuously monitored before and after injection of different doses of TMP at different times.The cranial window was made and the LDF probe was placed on the surface of the temporal lobe.Under the LDF successive monitoring,the ischemia model was made by occlusion of right middle cerebral artery with nylon suture,and the animals then received a single i.p.injection of TMP.Two hours after the occlusion,the nylon suture was withdrawn to allow reperfusion.
