1-methyl-4-phenylpyridinium ion (MPP+),an active metabolite of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP),induces apoptois in PC 12 cells.To study the mechanism whereby apoptosis is induced by MPP+,we treated the PC12 cells with MPP+.MPP+ induced the procaspase 12 decreased and the activated caspasel2 increased.Nerve growth factor (NGF) and glycogen synthase kinase-3b inhibitor LiCl restored the procaspase 12 after the MPP+ treatment in PC12 cells.As well as NGF and Licl suppressed the apoptosis induced by MPP+.These data suggest that MPP+ induced the apoptosis is through the endoplasmic reticular pathway and caspasel2 is the downstream of the Akt/glycogen synthase kinase-3 beta pathway.
