Characterization of the insecticidal activity of Pseudomonas taiwanensis toward Aedes albopictus lar

  The insecticidal activity of a novel Pseudomonas taiwanensis was analyzed against Aedes albopictus larvae.The P.taiwanensis were cultured in LB medium for 20 hr,afterward,the supernatant were collected for insecticidal bioassay.The results showed that the mosquito larvae were highly susceptible to the culture supernatant,but not the cell pellet,indicating that the insecticidal ingredient is a secretory product.The mortality of 2nd instar A.albopictus larvae could be higher than 80%after exposed to the cultural medium or supernatant for 2 hr and reach 100%mortality after 4 hr treatment.The 4th instar larvae exhibit less sensitivity toward P.taiwanensis,however it still showed around 50%mortality after 16 hr exposure to the supernatant.Though an insecticidal gene(TccC)toward Drosophila was identified and cloned from the P.taiwanensis(J.Agric.Food Chem.58,12343-12349(2010)),it is apparently not the responsible toxin for A.albopictus larvae,since the insecticidal activity is not affect by autoclave at 121℃for 30 min.Moreover,the supernatant of tccC knockout mutant showed even higher activity toward A.albopictus than parental P.taiwanensis.The supernatant of P.taiwanensis also exhibit similar toxic effect toward A.aegypti and Culex quinquefasciatus,but not other non-Culicidae.These results suggest that P.taiwanesis could be an ideal bioinsecticide specific for aquatic mosquito.
