Subcellular Distribution and Chemical Forms of Hg in Leaves of Kandelia obovata

  Foliage is the main organ for exchange of Hg between plant and atmosphere.Distribution and forms of Hg in leaves are of great significance to understand Hg absorption and release.In order to probe accumulation mechanism of Hg in Kandelia obovata—one of the widely distributed mangrove species in China,differential centrifugation,TEM and sequential chemical extraction methods were used to study sub-cellular distribution and chemical forms of Hg in the leaves of Kandelia obovata.Results were as follows: there was an important amount of Hg was found in the soluble fraction,in the second term,Hg was associated with cell-wall,followed by organelle containing fraction,while membrane containing fraction being the smallest.Accumulation content of Hg was enhanced with the increasing strength and time of Hg treatment.For the chemical forms,water extractable Hg predominated in leaves,residue was the second,which indicated that water soluble state of organic ligand may played a key role in the sub-cellular distribution of Hg.Vacuolar compartmentalizationand precipitation of cell walls was the main way for Hg enrichment of Kandelia obovata,which was an adaption strategy of Kandelia obovata for Hg.
