Much progress has been made over past several years in genetic reprogramming of T lymphocytes for fighting against cancer.We have now reached the point where we can achieve real and outstanding benefits for a significant fraction of cancer patients.Genetic manipulation and reverse engineering provide a distinct advantage for creating tumor-specific T cells exceeding the endogenous capacity of immune repertoire of patients.Synthetic engineering strategies can meet the essential requirements for an effective immune response against advanced cancer, which include enabling T cells to rec﹁ognize and respond against wide spread tumor cells, resisting to tumor imposed immunosuppression, as well as facilitating trafficking and homing into tumor bed and persistence for long periods of time.I will review the exciting new development in this field and discuss the opportunities we have and challenges we are facing to translate such major breakthrough into medical practice for cancer treatment in China.
