Objective To investigate whether genetic polymophisms in SLC17A7 gene is associated with serotonin reuptake inhibitors (SSRI) treatment response of major depressive disorder patients.Methods A total of 290 depressed patients, age 18-55 years, fulfilling the DSM-Ⅳ diagnostic criteria for major depressive disorder, with a minimum HAMD score at 18, were recruited for this study.All patients were randomly treated with 4 SSRIs and received a 6-week period of treatment.Response was defined as a >50% of the reducing scores rate of HAMD scores at the end of the 6th week.Four single nucleotide polymorphisms (SNPs) in SLC17A7 gene selected from either the HapMap Chinese ethnic group or the literature report were studied.Results There were two SNPs(rs1578944, rs74174284) associated with response to antidepressant treatment, but only rs74174284 remained significant after correcting for multiple testes using FDR method(FDR p=0.032).The haplotype C-C of rs74174284-rs1578944 showed less non-responder than responders (p=0.0086), while the haplotype G-T showed more non-responders than responders (p=0.0034).Conclusions Our results suggest that genetic variants in the SLC 17A7 gene may be indicators of treatment response in MDD patients treated by SSRIs.
