Pancreatic cancer, called the silent killer, is the fourth leading cause of cancer-related death in both men and women in US.Due to difficulties in diagnosis and resistance to chemotherapy as well as radiotherapy, pancreatic cancers five-year survival rate is only about 1% in US.Since emerging evidence suggests there are putative pancreatic cancer stem cells (PCSC), our research aims to develop new approaches based on molecular magnetic resonance imaging to image pancreatic cancer stem cells in vivo.We first conjugate magnetic nanoparticles with PCSC biomarker antibody, then incubate the antibody with PCSC in cell media.The binding between magnetic nanoparticles-antibody and PCSC is confirmed by an independent experiment: YAG (a fluorescence dye) pre-coated with lipofectamine was conjugated with EGF to form YAG-EGF nanoparticle, and then the YAG-EGF nanoparticles were bound to cells in cell media for imaging by light microscopy and fluorescence.After the binding is confirmed, we then image PCSC via magnetic resonance imaging with new pulse sequence for enhanced imaging contrast and sensitivity.
