Object To research the inhibiting effects of acteoside extracted chemical composition from cistanche tubulosa on experimental begnin prostatic hyperplasia (BPH) in rats.Methods The begnin prostatic hyperplasia rat model were induced by the castration combined injection of testosterone propionate, and two dose groups of acteoside (60, 20mg.kg-1.d-1) were administrated for 4 weeks, The Qianliekang used as positive control (540mg.kg-1.d-1), observed the morphological change of the prostate gland by the microscope.Exzamined androgen receptor (AR) and βtransformation growth factor Ⅰ recptor (TGFβ R1) expressions by the immunity histochemistry.Cell apoptosis by flow cytometry and morphological changes of prostate ultrastructure by scanning electron microscope.Results The acteoside could obviously reduced the prostate gland fresh weight index.Compared with the the model group, showed significant difference (p<0.01).Androgen receptor and β transformation growth factor Ⅰ recptor expressions reduced in the prostate gland epidermis cell nucleus in high dose group.Compared with the the model group,showed significant difference (p<0.01).Intervention by acteoside, rat prostate apoptosis was significantly higher than that of model group (p<0.01).Under scanning electron microscope observation, acteoside protected nucleus of prostate deformed and endoplasmic reticulum vacuolization significantly reduced than that of model group.Conclusion Acteoside could inhibited the rat begnin prostatic hyperplasia, by downing androgen receptor and β transformation growth factor Ⅰ recptor expression, inhibiting prostate epithelial cell proliferation and promoting prostate cells apoptosis.Its role better than Pule an Pian.
