Taxonomic statuses of three subterranean rodents,Lasiopodomys mandarinus,Myospalax psilurus and Myospalax baileyi are still in dispute.To clarify their status the complete mitochondrial genomes of these three rodents were sequenced.The gene content,organization of the mtDNA,and phylogenetic analysis based on 13 protein coding genes,2 rRNA genes and Cytb genes were constructed.The results were as follows: 1) the lengths of the complete mtDNA of L.mandarinus,M.psilurus and M.baileyi were 16367 bp,16351 bp,16360 bp respectively.These mtDNA have a typical gene content of 13 protein coding genes,22 tRNA genes and 2 rRNA genes,which are highly conserved among most rodents.2) Like other rodents,the base usage of the three mtDNA also follow the A+T>C+G rule.The codons usage,use of incomplete stop codons,tRNASer secondary structure lost its DHU arm,structures of D-loop and rep-origin are similar to other rodents mtDNA.3) Phylogenetic analysis with the support of morphological data showed that L.mandarinus has a close relationship with rodents in genus Microtus and Proedromys.Thus the Lasiopodomys should be retained as a separated genus of Cricetidae.Two zokors are species of Spalacidae,M.psilurus belongs to genus Myospalax,and M.baileyi belongs to genus Eospalax.The phylogenetic analysis also gave further evidence for the notion of Rodentia monophyly.In this study,mtDNA are efficient molecular markers for clarifying the taxonomic status of these subterranean rodents.
